CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. This antibody recognizes a protein of ~35 kDa, identified as CD74 (Workshop IV). CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74 stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas. Spectrin is an actin crosslinking and molecular scaffold protein that links the plasma membrane to the actin cytoskeleton, and functions in the determination of cell shape, arrangement of transmembrane proteins, and organization of organelles. Spectrins function as membrane organizers and stabilizers, composed of non-homologous α andβ chains, which aggregate side-to-side in an anti-parallel fashion to form dimers, tetramers and higher polymers. Thyroperoxidase (TPO) is a 933-amino acid, type I transmembrane glycoprotein that plays a key role in thyroid gland function and autoimmunity. It is present as a dimer on the apical surface of thyroid follicular cells. TPO functions in the iodination of tyrosine residues in thyroglobulin and phenoxy-ester formation between pairs of iodinated tyrosines to generate the thyroid hormones, thyroxine and triiodothyronine. Mutations in this gene are associated with several disorders of thyroid hormonogenesis, including congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA. Malignant thyroid tumors exhibit an anomaly in TPO resulting in lower affinity for anti-TPO. This antibody recognizes an intermediate filament protein (IFP) of 55 kDa, which is identified as cytokeratin 7. This MAb is highly specific to cytokeratin 7 and shows no cross-reaction with other IFPs. Cytokeratin 7 is a basic cytokeratin, which is found in most glandular and transitional epithelia but not in the stratified squamous epithelia. Keratin 7 is expressed in the epithelial cells of ovary, lung, and breast but not of colon, prostate, or gastrointestinal tract. Cytoskeletal intermediate filaments (IFs) constitute a diverse group of proteins that are expressed in a highly tissue-specific manner. IFs are constructed from two-chain -helical coiled-coil molecules arranged on an imperfect helical lattice, and have been widely used as markers for distinguishing individual cell types within a tissue and identifying the origins of metastatic tumors. Vimentin is an IF general marker of cells originating in the mesenchyme. Vimentin and Desmin, a related class III IF, are both expressed during skeletal muscle development. Desmin, a 469 amino acid protein found near the Z line in sarcomeres, is expressed more frequently in adult differentiated state tissues. Anti-desmin detects cells of normal smooth, skeletal, and cardiac muscles. Keratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I keratin (keratins 9 to 23) and one type II keratin (keratins 1 to 8). Keratins demonstrate tissue and differentiation specific expression profiles. Cytokeratin 1 is a hetero-tetramer of two type I as well as two type II keratins, keratin-10 (KRT10). Keratin 1 is differentiation specific keratin that is one of the predominant keratins in suprabasal keratinocytes in stratified epithelia. Mutations in keratin 1 cause epidermolytic hyperkeratosis. Caldesmon HMW is the high molecular weight variant of Caldesmon. Two closely related variants of human caldesmon have been identified which are different in their electrophoretic mobility and cellular distribution. The h-caldesmon variant (120'150 kDa) is predominantly expressed in smooth muscle whereas l-caldesmon (70'80 kDa) is found in non- muscle tissue and cells. Neither of the two variants has been detected in skeletal muscle. This antibody recognizes only the 150 kDa variant (h-caldesmon) in Western blots of human aortic media extracts and is unreactive with fibroblast extracts from cultivated human foreskin. Caldesmon is a developmentally regulated protein involved in smooth muscle and non-muscle contraction. von Willebrand Factor (vWF) is a multimeric glycoprotein that is found in endothelial cells, plasma and platelets. It acts as a carrier protein for Factor VIII and promotes platelet adhesion and aggregation. vWF undergoes a variety of posttranslational modifications that influence the affinity and availability for Factor VIII, including cleavage of the propeptide and formation of N-terminal disulfide bonds. This antibody helps to establish the endothelial nature of some lesions of disputed histogenesis, e.g. Kaposi's sarcoma and cardiac myxoma.
